A team of scientists in the UK developed non-invasive measurement techniques and novel analysis methods to decode disease progression and evaluate the effect of potential treatments or lifestyle changes in people with Huntington’s disease.
Huntington’s is a genetic disease where the brain’s nerve cells (neurons) gradually break down and die. The condition leads to dementia, with a progressive decline in an individual’s movement, memory, and cognition. There is currently no cure.
The team from Lancaster University in the UK showed that Huntington’s disease not only affects nerve cells in the brain but also has widespread effects on microscopic blood vessels.
The changes were also observed before symptoms of the disease appeared, demonstrating the potential for this research to predict brain health and evaluate the beneficial effects of lifestyle changes or treatments.
Professor Aneta Stefanovska of Lancaster University said that they hope that the novel method can help “monitor the disease progression and evaluate the effect of potential treatments or lifestyle changes” in people with “Huntington’s and other neurodegenerative diseases.”
Professor Aneta added that the study would also lead to “new treatments of Huntington’s disease targeting the vasculature and brain metabolism.”
In their study, published in the journal Brain Communications, the team investigated changes in the coordination between neuronal activity and the brain’s oxygenation in Huntington’s disease.

The US Supreme Court upholds access to a widely available abortion pill
WASHINGTON, D.C. (TIP): The Supreme Court on Thursday, June 13, maintained access to a widely available abortion pill, rejecting a bid from a group of anti-abortion organizations and doctors to undo the Food and Drug Administration’s approval of the drug.
In a unanimous decision, written by Justice Brett M. Kavanaugh, the court held that the anti-abortion groups lacked a direct stake in the dispute, a requirement to challenge the F.D.A.’s approval of the pill, mifepristone.
“The plaintiffs do not prescribe or use mifepristone,” Justice Kavanaugh wrote. “And F.D.A. is not requiring them to do or refrain from doing anything.”
He added, “A plaintiff ’s desire to make a drug less available for others does not establish standing to sue.”
The case originally sought to erase the F.D.A.’s approval of mifepristone. But by the time it reached the Supreme Court, the question had been narrowed to whether the agency had acted legally in 2016 and 2021, when it broadened distribution of the pill, eventually including telemedicine and mail options.
The ruling handed a muted victory to abortion rights groups. Even as they praised the decision for averting severe restrictions on the availability of the pill, they warned that the outcome could be short-lived. Anti-abortion groups vowed to press ahead, promising that the fight was far from over and raising the possibility that other plaintiffs, states in particular, would mount challenges to the drug.
The ruling did not affect separate restrictions on the pill in more than a dozen states that have passed near-total bans on abortion since the court eliminated a constitutional right to the procedure in Dobbs v. Jackson Women’s Health Organization. (The bans do not distinguish between medication and surgical abortion.)
Access to abortion remains broadly popular, and ever since the court overturned Roe v. Wade two years ago, prompting some states to swiftly enact bans, the issue has been a major focus of political campaigns. Democrats have succeeded in galvanizing voters to defeat anti-abortion measures and plan to highlight abortion rights in the November elections.
By dodging a ruling on the substance of the case, the justices avoided delivering a clear, substantive win to either political party or a decision they could use to motivate their base.
President Biden said in a statement that the “decision does not change the fact that the fight for reproductive freedom continues.”
He added, “It does not change the fact that the Supreme Court overturned Roe v. Wade two years ago, and women lost a fundamental freedom.”
The Biden campaign also raised concerns that the ruling would not be enough to protect access to abortion medication if former President Donald J. Trump wins a second term, saying his administration would move to enforce new restrictions through executive action.
Mr. Trump had wrestled with what position to take on abortion access after the Supreme Court, whose conservative supermajority he appointed, overturned the Roe v. Wade landmark abortion-rights case. Several weeks ago, he said it was up to states to set their own policies.
His campaign suggested he staked out a similar position on the abortion pill ruling, without addressing how his administration would handle regulation of the drug. “The Supreme Court has unanimously decided 9-0; the matter is settled,” Danielle Alvarez, a Trump spokeswoman, said.
Abortion rights groups cautioned that the ruling only maintained the status quo.
“The anti-abortion movement sees how critical abortion pills are in this post-Roe world, and they are hell bent on cutting off access,” Nancy Northup, the president of the Center for Reproductive Rights, said in a statement.
Erin Hawley, senior counsel for Alliance Defending Freedom, the conservative legal organization that represented the plaintiffs, suggested that the case could be revived through three Republican-led states, Idaho, Kansas and Missouri, which had intervened as plaintiffs at the lower court level.
“We are grateful that three states stand ready to hold the F.D.A. accountable for jeopardizing the health and safety of women and girls across this country,” Ms. Hawley said in a statement.
Mifepristone, part of a two-drug regimen, is used in nearly two-thirds of abortions in the United States. Many studies have found the pill to be safe, and years of research have shown that serious complications are rare.
After the F.D.A. loosened restrictions on the drug’s availability during the pandemic, allowing for it to be prescribed online or sent by mail, its use only increased. A rising number of medication abortions are prescribed by telemedicine. By one count, about one in six abortions, or around 14,000 a month, happened via telehealth from July through September 2023.
Medication abortion has become a practical form of abortion for women in states where the procedure is banned. Clinicians, protected by so-called shield laws in several states where abortion is legal, have mailed pills to women based in states with bans.
Justice Kavanaugh focused part of the majority opinion on why the justices disagreed with rulings by lower courts, which all determined the groups had standing to bring the case.
Citing Justice Antonin Scalia, Justice Kavanaugh wrote that to bring a suit, a plaintiff first must answer a basic question: “What’s it to you?”
Plaintiffs must show “a predictable chain of events leading from the government action to the asserted injury,” Justice Kavanaugh wrote.
In this instance, he wrote, the doctors and medical associations trying to challenge F.D.A.’s regulation failed to show an actual injury because the plaintiffs did not include people actually involved with the pill, such as doctors who prescribed mifepristone or pregnant women who took it.
The plaintiffs’ claims that they have “sincere legal, moral, ideological, and policy objections to mifepristone being prescribed and used by others” do not meet the threshold of standing to sue, Justice Kavanaugh wrote.
The justices also rejected an argument by the anti-abortion doctors that they had standing to sue because they could be required to provide emergency abortions against their conscience.
Federal conscience laws already protect doctors from being required to perform abortions or other treatment that violates their consciences, and none of the doctors had shown otherwise, Justice Kavanaugh wrote.
In a concurring opinion, Justice Clarence Thomas noted that he supported the decision “in full,” but cautioned that the court should take a limited view of when to allow organizations to claim standing on behalf of their members.
The case returned the issue of abortion to the Supreme Court, even as the conservative majority had declared, in overturning Roe v. Wade, that it would cede the question of access “to the people and their elected representatives.”
Its decision on Thursday appeared to vindicate that promise, although the court will soon decide another major case on abortion, involving a clash between federal law and tightened state restrictions.
The abortion pill case, F.D.A. v. Alliance for Hippocratic Medicine, reflected a new front in the fierce fight over abortion.
In the fall of 2022, an umbrella group of anti-abortion medical organizations, along with several doctors, challenged the F.D.A.’s approval of mifepristone more than two decades ago. In a preliminary ruling, a federal judge in Texas, Matthew J. Kacsmaryk, said that the F.D.A.’s approval of the drug should be suspended, removing mifepristone from the market.
Judge Kacsmaryk, a Trump appointee, joined the federal bench after years litigating at a conservative religious freedom firm, First Liberty Institute.
An appeals court, in New Orleans, overturned the part of Judge Kacsmaryk’s ruling that invalidated the F.D.A.’s approval of the pill, but it imposed certain restrictions on its distribution. Those restrictions included prohibiting the medication from being sent by mail or prescribed by telemedicine.

Alcohol-based mouthwash may not be good for your oral health: Study
Commonly used alcohol-based mouthwashes may have adverse effects on oral microbiomes—the community of bacteria that live in the mouth—raising risks of periodontal diseases and some cancers, according to a study.
The oral microbiome plays a role in digestion and keeps the mouth healthy.
The research, published in the Journal of Medical Microbiology, included men who have sex with men, who regularly use mouthwash as a method to reduce the transmission of sexually transmitted diseases.
Three months of daily use of the alcohol-based mouthwash increased the amount of two species of opportunistic bacteria—Fusobacterium nucleatum and Streptococcus anginosus—in the mouth of these men, said the team from the Institute of Tropical Medicine in Antwerp in Belgium.
The two bacteria are known to raise the risk of gum disease, and oesophageal and colorectal cancers.
Further, the researchers also observed a decrease in a bacteria group called Actinobacteria—crucial for regulating blood pressure.
“Alcohol-based mouthwashes are widely available. The public may use them daily to tackle bad breath or prevent periodontitis, but they should be aware of the potential implications. Ideally, long-term usage should be guided by healthcare professionals,” said Jolein Laumen, from ITM’s Unit of Sexually Transmitted Infections.

Low-dose aspirin eases inflammation caused by sleep loss: Study
A recent study discovered that low-dose acetylsalicylic acid, popularly known as aspirin, can attenuate inflammatory responses to sleep deprivation. Study is to be presented at the SLEEP 2024 annual meeting.
The results suggest that prior treatment of low-dose aspirin during sleep restriction lowered pro-inflammatory responses compared to a placebo. Aspirin specifically reduced interleukin-6 expression, COX-1/COX-2 double positive cells in lipopolysaccharide-stimulated monocytes, and C-reactive protein serum levels.
“The novelty of this study is that it investigated whether we can pharmacologically reduce the inflammatory consequences of sleep restriction,” said lead author Larissa Engert, who has a doctorate in behavioral physiology and is a postdoctoral fellow in the department of neurology at Beth Israel Deaconess Medical Center and the division of sleep medicine at Harvard Medical School in Boston. “We used a non-steroidal, anti-inflammatory drug because it has been shown to affect specific inflammatory pathways, which were previously shown to be dysregulated by experimental sleep restriction or sleep disturbances.”
The researchers collected data from 46 healthy adults in a randomized placebo-controlled crossover trial with three protocols – sleep restriction/aspirin, sleep restriction/placebo, and control sleep/placebo – each consisting of a 14-day at-home phase followed by an 11-day in-hospital stay. In the sleep restriction/aspirin condition, participants took low-dose aspirin during the at-home phase and in-hospital stay. Each in-hospital stay started with two nights of an eight-hour sleep opportunity. Then, under the sleep restriction conditions, participants were exposed to five nights of a four-hour sleep opportunity, followed by three nights of recovery sleep. The control sleep condition provided an eight-hour sleep opportunity throughout the in-hospital stay. Sleep and immunologic measures were assessed at baseline and various points throughout the study. Statin therapy can reduce heart disease and death in elderly
Using statin therapy as the first line of treatment for cardiovascular disease (CVD) prevention was found effective in preventing heart disease risk and all-cause mortality, even in adults aged 85 years and older, according to a study on Tuesday. Statins are known to lower cholesterol levels in the blood, thereby decreasing the risk of CVDs.
Previously, the consensus on using statins for primary prevention of heart disease in adults aged 75 years or older has been little because of a lack of proper representation of the population in randomised controlled trials, said researchers from the University of Hong Kong.
The study, published in the Annals of Internal Medicine, included adult patients over 60 without pre-existing diagnosed CVDs. The average follow-up was 5.3 years.
Of the 42,680 adults aged 75 to 84 years, 9,676 developed CVDs. Among the 5,390 adults aged 85 years or older, 1,600 developed CVD.
The results showed that in all age groups, initiating statin therapy was associated with a lower incidence of CVD and all-cause mortality, said the researchers.
This was seen “even among the older population aged 85 years or older,” they added.
The new study showed that statin use did not increase the risk for adverse events such as myopathies and liver dysfunction.

Why depression, anxiety is common among children with ADHD?
A smaller brain region responsible for control over behaviour, processing of emotion, and communication may explain why about one in two children with Attention Deficit Hyperactivity Disorder (ADHD) is likely to experience depression, anxiety, and explosive outbursts, according to a study.
ADHD affects around one in 14 young people under the age of 18 and in around half of these cases it persists into adulthood. Mood disorders such as depression, anxiety disorder, and verbal or physical outbursts due to an inability to regulate their emotions in children with ADHD were previously thought to be a result of problems with cognition and motivation.
But in the study, scientists from Fudan University in Shanghai in China and the University of Cambridge showed that emotional dysregulation occurs independently of these, and is because of a brain region called pars orbitalis.
“The pars orbitalis is a well-connected part of the brain, and if it hasn’t developed properly it might make it difficult for individuals to control their emotions and communicate with others appropriately, especially in social situations,” said Barbara Sahakian from the Department of Psychiatry at the University of Cambridge. For the study, published in the journal Nature Mental Health, the team identified 350 individuals with high symptoms of ADHD and found that more than half (51.4 per cent) had signs of emotion dysregulation and this was independent of cognitive and motivational problems.
Children with only low-ADHD symptoms but with a high score of emotion dysregulation at age 13 years were 2.85 times more likely to have developed high-ADHD symptoms by age 14 years.

To put this into perspective, a 300-calorie mango accounts for a substantial portion of the recommended daily calorie intake for many individuals. If an average adult requires around 2,000 calories per day, a single large mango would contribute roughly 15% of their total daily calorie needs,” adds Kapoor.

New Covid variant ‘FLiRT’ capable of evading the immune system: Experts
The new Covid-19 variant ‘FLiRT’, which is spreading fast in the US, is capable of evading the immune system due to a trans substitution of amino acids between two spike proteins, the experts have said.
FLiRT, which belongs to Omicron’s JN1 lineage, is rapidly replacing the previous variant, Eris, in the US, the UK, New Zealand, and South Korea, according to Rajeev Gupta, Director of Internal Medicine at the CK Birla Hospital® in Delhi.
“A recent increase in hospitalisation rates in these countries has been attributed to this variant. However, it has remained a relatively small wave. The overall mortality rate has not increased,” Gupta said. According to the Infectious Disease Society of America, the nickname ‘FLiRT’ is based on the technical names for their mutations.
The World Health Organisation has reclassified it as a variant of interest and advised close monitoring. According to Dhiren Gupta, a Pediatric Intensivist at Sir Ganga Ram Hospital, these new strains will keep emerging.
“Fortunately, none of the Omicron lineage is able to induce significant lung damage as delta strain did but limited to the upper respiratory tract. The surveillance and vigilance should be kept for major drift in virus,” he said.
According to the experts, the increased use of monoclonal antibodies could be driving this mutation.
The symptoms of the new variant are similar to those of other Omicron sub-variants, such as sore throat, cough, fatigue, nasal congestion, runny nose, headache, muscle aches, fever, and possible loss of taste and smell, the experts mentioned. -

Repurposed cancer drug to help replace insulin therapy for diabetes
A drug used for the treatment of pancreatic cancer can be repurposed to replace insulin therapy among people with diabetes, scientists have found. Without enough insulin, diabetics could be at risk for hyperglycemia, or high blood sugar that can damage blood vessels and organs as well as cause heart attacks, strokes, and other serious complications.
The study, published in the journal Nature Communications, suggested that Focal Adhesion Kinase (FAK) inhibitors, known to reduce tumour burden in pancreatic cancer, could be a new avenue as a replacement for insulin therapy in diabetic patients.
In an experiment in mice that started in 2016, the team from the University of Pittsburgh deleted one of two copies of the gene encoding an enzyme called FAK.
Both the pancreas as well as a cluster of cells in the organ looked weird. While the pancreas looked “like it was trying to regenerate after an injury”, the cells were “expressing both insulin and amylase”.
The cluster of cells looked like a combination of acinar cells—that manufacture amylase, a digestive enzyme, and beta-cells—which produce the blood sugar-regulating hormone insulin.
“There were three possible explanations for what we saw in the mutant mice,” said Esni. “It could have just been an artefact of our experiment, beta cells could have started making amylase or acinar cells could have started producing insulin—which would be the holy grail.”
The team further showed that a “FAK-inhibiting drug, which has been studied in cancer treatment, converted acinar cells into acinar-derived insulin-producing cells and helped regulate blood glucose in diabetic mice and a single non-human primate”. -

Meta-owned WhatsApp is reportedly testing a new feature that will offer users a dedicated “filter” to quickly get a list of their favourites from the chats tab on Android.
According to WABetaInfo, the feature is set for release in a future update of the app.
With this new chat filter, users will get to easily access and prioritise specific conversations with their favourite contacts and groups.
As per the report, this new tool will let users mark specific contacts and groups as favourites, making it easier to prioritise important chats and access those they engage with most often.
This feature is presently under development and it will be available in a future update of the app, the report added.
Meanwhile, WhatsApp is reportedly working on a new feature that will let users react quickly to status updates on Android.
With the ‘quick reaction feature for status updates’, users will be able to quickly add a reaction to status updates, allowing them to express their appreciation for the posted content with ease.
